Lucky Star zeigt den Alltag der vier Highschool Schülerinnen Konata, Kagami, Tsukasa und Miyuki, sowie deren Freunde und Familie. In ihren Erlebnissen und Gesprächen über Gott und die Welt werden die diversen Kuriositäten der japanischen Kultur aufgezeigt. Besonders die Subkultur der Otakus wird hier beleuchtet, vertreten durch Konata, die an wenig Anderem interessiert ist und deren Schulleistungen unter ihrem exzessiven Anime- und Videospielkonsum leiden, was von Kagami laufend mit sarkastischen Sprüchen kommentiert wird.
